Exression and clinical sinificance of HBx and Beclin1 in heatocellular carcinoma

Abstract: Objective To investigate the specific role of hepatitis B virus X protein (HBx)and Beclin1 protein(Beclin1)in the pathogenesis of hepatocellular carcinoma(HCC)and their correlations.Methods Carcinoma tissue and adjacent tissue from 64 patients pathologically diagnosed as HCC without any treatment before surgery were collected in our hospital from January 2014 to December 2015.The expressions of HBx and Beclin l in carcinoma tissue and adjacent tissue of all cases were detected by real-time polymerase chain reaction(PCR),western blot and immunohistochemistry(IHC),which were analyzed in detail combined with their clinical data.Results Compared to those in adjacent tissue,higher levels of HBx and Beclin1 were detected by IHC in 82.8%and 79.7%of HCC tissue,respectively.Meanwhile,both HBx and Beclin1 showed higher protein levels in HCC tissues than that in adjacent tissues by western blot,as well as m RNA levels(0.78±0.10 vs.0.11±0.01 and 0.83±0.15 vs.0.18±0.03)by real-time PCR,respectively.Conclusion HBx and Beclin1 are both overexpressed in HCC and might participate in the pathogenesis of HCC.Combined detection of HBx and Beclin1 might play an important role in prediction of occurrence and metastasis of HCC,which might be potential target genes for HCC treatment.
